Belzutifan (PT2977) is an orally active, selective hypoxia-inducible factor-2α (HIF-2α) inhibitor. By selectively binding to the PAS-B domain of HIF-2α, it blocks heterodimerization with HIF-1β, preventing the assembly of a functional transcriptional complex. Disruption of this complex downregulates downstream target gene expression, including vascular endothelial growth factor (VEGF), which subsequently suppresses tumor cell proliferation, angiogenesis, and survival in clear cell renal cell carcinoma models.
