BAY 2402234 (Orludodstat) is a potent, selective dihydroorotate dehydrogenase (DHODH) inhibitor with an IC50 of. By blocking DHODH activity within the mitochondrial inner membrane, it disrupts the de novo pyrimidine biosynthesis pathway, causing depletion of intracellular pyrimidine pools. This metabolic inhibition induces cell differentiation, suppresses cell proliferation, and triggers apoptosis in myeloid leukemia cells, effectively impairing tumor growth in preclinical models. [1]
