BAY 1000394 (Roniciclib) is a potent pan-CDK inhibitor targeting CDK1 (IC50 = 7 nM), CDK2 (IC50 = 9 nM), CDK4 (IC50 =, CDK7 (IC50 = 25 nM), and CDK9 (IC50 = 5 nM). By inhibiting cyclin-dependent kinase activity, it disrupts cell-cycle progression and transcriptional regulation, thereby triggering cell-cycle arrest and apoptosis in tumor cells and exhibiting strong preclinical antitumor efficacy. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
