Siponimod (BAF312) is a selective sphingosine-1-phosphate receptor agonist targeting S1P1 and S1P5, displaying over 1000-fold selectivity over S1P2, S1P3, and S1P4 receptors. Binding to S1P1 induces receptor internalization and inhibits lymphocyte egress from secondary lymphoid tissue into circulation. This receptor modulation suppresses downstream neuroinflammatory pathways and reduces pathogenic autoimmune cell infiltration. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
