Biological Activity
Baclofen is a selective gamma-aminobutyric acid type B (GABAB) receptor agonist that acts as a skeletal muscle relaxant and central nervous system depressant. Activation of transmembrane GABAB receptors by baclofen suppresses presynaptic neurotransmitter release through the modulation of inward-rectifying potassium channels and voltage-gated calcium channels. This downstream signal transduction decreases spinal monosynaptic and polysynaptic reflex transmission, leading to reduced muscle tone and spasticity. [1][2]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
