b-AP15 is a potent small-molecule inhibitor of 19S proteasome deubiquitinase activity, blocking proteasomal cleavage of Ub-AMC with an IC50 of. By inhibiting the 19S regulatory particle enzymes USP14 and UCHL5, b-AP15 impairs proteasomal protein degradation. This inhibition results in the rapid accumulation of high-molecular-weight polyubiquitinated proteins, triggering severe endoplasmic reticulum stress and inducing caspase-dependent apoptosis in various cancer cell models. - Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
Pre-aliquoted for Longevity: Solid-state aliquots offer significantly longer shelf life compared to stock solutions, ensuring maximum stability for every use.
