Azosemide is a loop diuretic and inhibitor of Na+–K+–2Cl− cotransporter NKCC1 variants hNKCC1A (IC50 0.246 µM) and NKCC1B (IC50 0.197 µM). By suppressing NKCC1-mediated ion transport, it disrupts intracellular chloride homeostasis in neuronal and peripheral tissues. This inhibition alters neuronal excitability and transepithelial salt secretion, downstream affecting cell volume regulation and intracellular ion dynamics. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
