Biological Activity

AZ628 is a pan-Raf inhibitor targeting CRAF, BRAF V600E, and wild-type BRAF, while also inhibiting VEGFR2, DDR2, Lyn, Flt1, and FMS. By suppressing the MAPK/ERK signaling pathway, AZ628 inhibits cell growth, induces cell cycle arrest, and triggers apoptosis in BRAF V600E-mutated cancer cell lines. Upregulated CRAF expression confers acquired resistance to AZ628 upon continuous exposure. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
