Biological Activity

AZ20 is a potent and selective small-molecule inhibitor of ATR kinase (IC50 5 nM) that demonstrates 8-fold selectivity over mTOR. Inhibition of ATR kinase activity by AZ20 blocks DNA damage checkpoint signaling, decreases CHK1 phosphorylation, and impairs homologous recombination repair. Consequently, treatment with AZ20 increases DNA double-strand breaks, inducing robust cell cycle arrest and apoptosis in replication-stressed cancer cell lines. [1]

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
