Spebrutinib (AVL-292) is a covalent, orally active, and highly selective Bruton's tyrosine kinase (BTK) inhibitor (IC50 <. It selectively targets Cys481 of BTK to block B-cell receptor (BCR) signaling and downstream activation of the NF-kB pathway. Consequently, this inhibition reduces pro-inflammatory cytokine production, impairs osteoclast differentiation, and suppresses disease severity in preclinical animal models of rheumatoid arthritis. A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
