Avanafil is a highly selective inhibitor of phosphodiesterase 5 (PDE5) with an IC50 of, demonstrating over 121-fold selectivity for PDE5 compared to other PDE isoenzymes. By blocking PDE5-mediated hydrolysis of cyclic guanosine monophosphate (cGMP), it elevates intracellular cGMP concentrations and activates cGMP-dependent protein kinase downstream signaling. Consequently, this pathway activation promotes vascular smooth muscle relaxation and enhances tissue vasodilation. [1]
