Xevinapant (AT406) is an orally active inhibitor of apoptosis protein (IAP) antagonist targeting cIAP1 (Ki 1.9 nM), cIAP2 (Ki 5.1 nM), and XIAP (Ki 66.4 nM). By mimicking the Smac AVPI peptide, it antagonizes XIAP to restore caspase-9 activity and induces rapid ubiquitination and degradation of cIAP1. Consequently, xevinapant promotes caspase cascade activation, triggers apoptosis, and suppresses tumor growth in preclinical human cancer models. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
Pre-aliquoted for Longevity: Solid-state aliquots offer significantly longer shelf life compared to stock solutions, ensuring maximum stability for every use.
