Biological Activity

Amifostine trihydrate is a broad-spectrum cytoprotective agent and radioprotectant. As a prodrug, it is dephosphorylated by cell-surface alkaline phosphatase to its active free thiol metabolite, WR-1065. WR-1065 directly scavenges reactive oxygen species and inactivates toxic chemotherapeutic metabolites. Through this ROS-scavenging mechanism, amifostine attenuates oxidative stress, reduces DNA damage, and prevents apoptosis in normal tissues during cisplatin chemotherapy and radiation therapy. [1]

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
