Biological Activity

AMG-9810 is a potent, competitive vanilloid receptor 1 (TRPV1) antagonist that inhibits capsaicin-induced activation of human TRPV1 (IC50 24.5 nM) and rat TRPV1 (IC50 85.6 nM). It blocks multiple modes of TRPV1 activation, including protons, heat, and endogenous ligands such as anandamide. By blocking TRPV1-mediated ion influx, AMG-9810 modulates downstream pain signaling and inflammatory pathways in preclinical pain models. [1]

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
