Biological Activity

AMG 193 is an orally active, MTA-cooperative inhibitor of protein arginine methyltransferase 5 (PRMT5) that selectively targets MTAP-deficient tumor cells over normal wild-type cells. By preferentially binding PRMT5 in the presence of methylthioadenosine, it suppresses methyltransferase activity to induce DNA damage, cell cycle arrest, and aberrant alternative mRNA splicing. Consequently, AMG 193 inhibits cell proliferation with approximately 40-fold selectivity and drives robust antitumor efficacy in preclinical xenograft models. [1][2]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
