AGI-6780 is a potent and selective inhibitor of the mutant enzyme IDH2 R140Q. By targeting the IDH2 R140Q mutant protein, it blocks the aberrant enzymatic activity responsible for converting alpha-ketoglutarate to the oncometabolite 2-hydroxyglutarate (2-HG). Lowering 2-HG levels relieves blockades in cell differentiation and induces lineage maturation in mutant IDH2 acute myeloid leukemia cells. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
