Biological Activity

Adavosertib (AZD1775, MK-1775) is a potent and selective Wee1 kinase inhibitor with an IC50 of 5.2 nM. It inhibits Wee1-mediated phosphorylation of CDK1 at Tyr15, thereby abrogating the G2 DNA damage checkpoint and promoting cell cycle progression into premature mitosis. Consequently, adavosertib induces apoptosis and sensitizes p53-deficient tumor cells to DNA-damaging agents.

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
