Acesulfame potassium is a non-nutritive sweetener that functions as an agonist of the human sweet taste receptor heterodimer T1R2/T1R3 and specific bitter taste receptors such as TAS2R4 and TAS2R43. Ligand binding to these G-protein–coupled receptors triggers downstream intracellular calcium mobilization and gustatory signal transduction in taste cells, giving rise to sweet taste perception along with secondary bitter taste responses. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
