Acemetacin is a non-steroidal anti-inflammatory drug and a carboxymethyl ester prodrug of indomethacin that targets cyclooxygenase (COX). Upon metabolic conversion to indomethacin, it inhibits COX-mediated arachidonic acid metabolism, suppressing prostaglandin synthesis. Downstream of COX inhibition, acemetacin reduces inflammatory mediator production, cell recruitment, and tissue inflammation, while demonstrating lower gastric mucosal toxicity compared to indomethacin.
