ABTL-0812 (α-Hydroxylinoleic acid) is a first-in-class orally available antitumor compound that inhibits the AKT/mTOR pathway. It functions by inducing the overexpression of TRIB3, an endogenous inhibitor of AKT, leading to the inhibition of AKT phosphorylation and subsequent inactivation of mTORC1. This pathway suppression triggers sustained cytotoxic autophagy and inhibits cell proliferation. ABTL-0812 also induces AMPK activation and ROS accumulation. [1]
