ZSTK474 is a pan-class I PI3K inhibitor targeting PI3Kγ (IC50 6 nM), PI3Kα (IC50 17 nM), PI3Kδ (IC50 37 nM), and PI3Kβ (IC50 53 nM). By inhibiting class I phosphatidylinositol 3-kinases, it attenuates downstream AKT/mTOR signaling pathways, resulting in cell cycle arrest and cellular growth inhibition. In animal models, oral administration of ZSTK474 exerts significant antitumor activity against human cancer xenografts with manageable toxicity.

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
