Belnacasan (VX-765) is an orally bioavailable prodrug of VRT-043198, an irreversible inhibitor of caspase-1. Upon conversion to its active metabolite VRT-043198, it selectively blocks caspase-1 activity, thereby preventing the cleavage and secretion of pro-inflammatory cytokines such as IL-1β and IL-18. This inhibition attenuates downstream inflammatory pathways, reducing disease severity in models of rheumatoid arthritis and suppressing acute partial seizures. [1][2]

- Preheat media/saline to 37°C.
