U0126-EtOH is a highly selective inhibitor of MEK1 (IC50 of and MEK2 (IC50 of. It functionally antagonizes AP-1 transcriptional activity through noncompetitive inhibition of dual-specificity MEK kinases, thereby blocking downstream MAPK/ERK signaling pathway activation. Consequently, this kinase inhibition suppresses anchorage-independent cell growth, blocks cellular proliferation, and induces apoptosis in constitutively active ERK-dependent cancer cell lines. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
