Biological Activity

NSC 74859 (S3I-201) is a chemical probe inhibitor of STAT3 with an IC50 offor STAT3 DNA-binding activity in cell-free assays, displaying low activity against STAT1 and STAT5. It disrupts STAT3-STAT3 complex formation and blocks STAT3-mediated transcriptional activity. Consequently, NSC 74859 downregulates STAT3 target genes including cyclin D1, Bcl-xL, and survivin, suppressing cell proliferation, inducing apoptosis, and inhibiting human breast tumor growth in vivo. FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
