R406 (free base) is an orally active spleen tyrosine kinase (Syk) inhibitor with a Ki of 30 nM (cell-free IC50 of. Downstream of Syk suppression, R406 blocks Fc receptor-mediated activation in monocytes, macrophages, and neutrophils, as well as B-cell receptor (BCR) signaling in B lymphocytes. Consequently, it inhibits immune complex-mediated inflammatory cascades and triggers apoptosis in target cells. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
