Pyrazinamide is a key antitubercular prodrug converted by the mycobacterial pyrazinamidase enzyme into its active metabolite, pyrazinoic acid. Accumulation of pyrazinoic acid disrupts bacterial membrane potential, inhibits trans-translation through binding to ribosomal protein S1, and impairs energy metabolism in Mycobacterium tuberculosis, ultimately leading to effective depletion of intracellular ATP and inhibition of bacterial growth under acidic conditions. - Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
Pre-aliquoted for Longevity: Solid-state aliquots offer significantly longer shelf life compared to stock solutions, ensuring maximum stability for every use.
