Biological Activity

Pemetrexed disodium is a novel antifolate and antimetabolite that functions as a multi-targeted inhibitor of thymidylate synthase (TS, Ki 1.3 nM), dihydrofolate reductase (DHFR, Ki 7.2 nM), and glycinamide ribonucleotide formyltransferase (GARFT, Ki 65 nM). By suppressing these essential folate-dependent enzymes, pemetrexed disodium disrupts nucleotide biosynthesis required for DNA and RNA synthesis, ultimately inducing cell cycle arrest, autophagy, and apoptosis in tumor cells. A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
