Biological Activity

Pamidronate Disodium is a nitrogen-containing bisphosphonate that acts as an inhibitor of farnesyl pyrophosphate synthase (FPPS) within the mevalonate signaling pathway. By impairing FPPS activity, pamidronate prevents the post-translational prenylation of essential small GTP-binding proteins, such as Ras, Rho, and Rac. Consequently, this disruption causes cytoskeletal reorganization, loss of ruffled border formation, and induction of apoptosis in osteoclasts, ultimately suppressing bone resorption. [1][2]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
