Biological Activity

Abexinostat (PCI-24781) is a novel pan-histone deacetylase (HDAC) inhibitor targeting HDAC1 with a Ki of, displaying modest potency against HDAC2, HDAC3, HDAC6, and HDAC10 while showing greater than 40-fold selectivity over HDAC8. By inhibiting HDAC enzymes, Abexinostat restores histone acetylation, suppresses downstream oncogenic pathways, and promotes cell cycle arrest and apoptosis. It exhibits preclinical broad-spectrum antitumor activity across multiple cancer cell lines and reduces tumor burden in xenograft models. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
