Org 27569 is an allosteric modulator of the CB1 cannabinoid receptor. It enhances the binding affinity of the agonist CP 55,940 (pKB = 5.67) while decreasing the binding of the inverse agonist SR141716A (pKB = 5.95). Functionally, Org 27569 acts as an insurmountable antagonist of CP 55,940-stimulated receptor activation (pKB = 7.57 in mouse vas deferens). Downstream, it inhibits agonist-induced CB1 signaling pathways and regulates receptor trafficking. [1]
