Nimesulide is a selective cyclooxygenase-2 (COX-2) inhibitor that targets prostaglandin-endoperoxide synthase 2 to block prostaglandin E2 synthesis. By inhibiting COX-2 activity, nimesulide disrupts downstream arachidonic acid metabolism and attenuates pro-inflammatory signaling pathways, including the NF-kB pathway. Consequently, this suppression impairs inflammatory mediator release, reduces cell proliferation, and promotes apoptosis in various cancer and inflammatory cell models. [1]
