Biological Activity

Luminespib (NVP-AUY922) is a potent heat shock protein 90 (HSP90) inhibitor targeting HSP90α and HSP90β (IC50 21 nM). By blocking the N-terminal ATP-binding domain of HSP90, Luminespib disrupts molecular chaperone activity and destabilizes oncogenic client proteins, including IGF-1Rβ. Consequently, this target inhibition impairs downstream cell survival pathways, leading to tumor cell growth inhibition, autophagy, and induction of apoptosis across multiple cancer models. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
