Biological Activity

Plinabulin (NPI-2358) is a potent vascular disrupting agent that targets the colchicine-binding site of tubulin, exhibiting an IC50 of 15 nM in HT-29 tumor cells. By inhibiting tubulin polymerization and disrupting tubulin dimerization, it destabilizes the microtubule network in proliferating endothelial cells. This leads to rapid endothelial cell shape changes, increased monolayer permeability, selective collapse of tumor vasculature, and induction of tumor cell apoptosis.

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
