Manidipine is a third-generation dihydropyridine calcium channel antagonist that acts as a potent, vasoselective inhibitor of voltage-dependent calcium currents. By blocking voltage-gated calcium influx in vascular smooth muscle cells, manidipine disrupts calcium-dependent signaling and excitation-contraction coupling. This inhibition reduces vascular resistance and induces systemic vasodilation, while demonstrating minimal cardiac inotropic suppression. [1]
