Biological Activity

MLN2238 (Ixazomib) is a reversible, selective 20S proteasome inhibitor targeting the β5 subunit,, with activity against the β1 and β2 subunits. By blocking proteasomal cleavage, it disrupts protein homeostasis, induces autophagy, and triggers cell cycle arrest and apoptosis. MLN2238 demonstrates potent antitumor efficacy in preclinical models of hematologic malignancies, including bortezomib-resistant lymphoma. [1][2]

A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
