Biological Activity

Leucovorin (folinic acid) calcium pentahydrate is a reduced folate derivative and biochemical modulator that targets the folate metabolic pathway. As an active metabolite precursor, it expands intracellular 5,10-methylenetetrahydrofolate pools, stabilizing the inhibitory ternary complex with thymidylate synthase and fluorodeoxyuridylate to enhance cytotoxicity. Additionally, leucovorin circumvents dihydrofolate reductase inhibition, rescuing non-malignant and lymphoid cells from methotrexate-induced growth suppression and restoring purine and pyrimidine biosynthesis. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
