Leflunomide is a disease-modifying antirheumatic drug that acts as a pyrimidine synthesis inhibitor, protein tyrosine kinase inhibitor, and aryl hydrocarbon receptor (AhR) agonist. Its active metabolite, A77 1726, potently inhibits dihydroorotate dehydrogenase (DHODH), a crucial enzyme in de novo pyrimidine biosynthesis. This inhibition suppresses pyrimidine nucleotide production, leading to cell cycle arrest and inhibition of activated immune cell proliferation, thereby conferring immunosuppressive activity. [1]

- Preheat media/saline to 37°C.
