Biological Activity

Varespladib (LY315920) is a potent and selective inhibitor of human group IIA secretory phospholipase A2 (sPLA2) with an IC50 of 9.6 nM. It exhibits 40-fold selectivity over human group IB pancreatic sPLA2 and does not inhibit cytosolic PLA2 or cyclooxygenase enzymes. By blocking sPLA2-mediated enzymatic activity, Varespladib suppresses the downstream release of inflammatory lipid mediators, including thromboxane A2, thereby exerting strong anti-inflammatory physiological effects.

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
