LY2811376 is an orally available, non-peptidic β-secretase (BACE1) inhibitor with an IC50 range ofand an EC50 offor decreasing amyloid-β secretion. It displays 10-fold selectivity for BACE1 over BACE2 and over 50-fold selectivity over other aspartic proteases, including cathepsin D, pepsin, and renin. Inhibition of BACE1 suppresses β-amyloid precursor protein cleavage, leading to robust reduction of amyloid-β production. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
