Biological Activity

Serdemetan (JNJ-26854165) is an orally bioavailable antagonist of the human double minute 2 (HDM2) ubiquitin ligase. By disrupting the interaction between HDM2 and p53, it prevents the proteasomal degradation of p53, thereby activating downstream p53 signaling pathways. This blockade restores p53-mediated transcription, suppresses cellular proliferation, and induces apoptosis in p53 wild-type tumor cells while also promoting delayed apoptotic responses in p53-deficient models. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
