Biological Activity

Ispinesib (SB-715992) is a potent and selective inhibitor of kinesin spindle protein (KSP/Eg5) with a Ki ofand an IC50 of. By inhibiting KSP ATPase activity, it disrupts bipolar mitotic spindle assembly, triggering cell cycle arrest in the M phase and inducing apoptotic cell death across various cancer cell lines. In vivo, ispinesib exhibits broad antitumor activity in human tumor xenograft models.

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
