GSK461364 is an ATP-competitive polo-like kinase 1 (PLK1) inhibitor (Ki 2.2 nM). It exhibits over 1000-fold selectivity for PLK1 compared to PLK2 and PLK3. Inhibition of PLK1 disrupts mitotic spindle assembly and chromosome alignment, leading to G2/M cell cycle arrest, inhibition of cancer cell proliferation, and induction of apoptosis across various tumor cell lines.

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
