Biological Activity

Erlotinib Hydrochloride is a potent EGFR inhibitor with an IC50 of 2 nM in cell-free assays and shows over 1000-fold selectivity for EGFR over c-Src and v-Abl. It inhibits EGFR autophosphorylation in intact tumor cells with an IC50 of 20 nM. By blocking EGFR kinase activity, erlotinib induces p27 accumulation and downregulates cyclin D1 expression, leading to cell-cycle arrest at the G1 phase and inhibition of cell proliferation. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
