Mechanism and Biochemical Profile

Enzastaurin selectively binds to and inhibits protein kinase C beta (PKCβ), which blocks downstream phosphorylation of key signaling components such as GSK3β, AKT, and ribosomal protein S6. This disruption suppresses tumor cell proliferation, induces apoptosis, and impairs microvessel density and angiogenesis, providing the mechanistic rationale for its clinical evaluation in conditions such as diffuse large B-cell lymphoma, glioblastoma multiforme, and advanced solid tumors.
