Dofetilide is a selective class III antiarrhythmic agent that acts as a potent blocker of the rapid component of the delayed rectifier potassium current (IKr), encoded by the hERG gene. By blocking hERG/IKr channels, dofetilide prolongs the cardiac action potential duration and effective refractory period without altering conduction velocity. These effects inhibit abnormal cardiac automaticity and re-entry pathways, suppressing atrial and ventricular arrhythmias. [1][2]

- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
