Diphenhydramine HCl is a classic first-generation histamine H1 receptor antagonist that competitively blocks histamine binding to H1 receptors. By inhibiting H1 receptor-mediated signal transduction, it suppresses histamine-induced phosphoinositide hydrolysis, intracellular calcium mobilization, and downstream NF-kB pathway activation. Consequently, this competitive antagonism reduces histamine-driven cellular responses, vascular permeability, and inflammatory tissue phenotypes. [1]

- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
