Biological Activity

Celastrol (Tripterine) is a pentacyclic triterpene that acts as a potent proteasome inhibitor, targeting the chymotrypsin-like activity of the purified 20S proteasome with an IC50 of 2.5 μM. By disrupting proteasomal degradation, it modulates ROS/JNK signaling pathways to induce cell cycle arrest, apoptosis, and autophagy. Furthermore, Celastrol promotes mitophagy to protect dopaminergic neurons and suppresses inflammatory cytokine release in immune cells. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
