Bumetanide is a loop diuretic that acts as a selective inhibitor of the Na+-K+-2Cl- cotransporter 1 (NKCC1) and weakly inhibits NKCC2. By suppressing NKCC-mediated transmembrane transport of sodium, potassium, and chloride ions, bumetanide disrupts intracellular ion homeostasis and osmotic balance. This inhibition impairs downstream intracellular chloride accumulation and cell volume regulation, leading to altered cellular excitability and electrolyte transport mechanisms. [1]

- Preheat media/saline to 37°C.
