Biological Activity

Brivanib (BMS-540215) is an ATP-competitive tyrosine kinase inhibitor targeting VEGFR-2 (IC50 25 nM), VEGFR-1 (IC50 380 nM), VEGFR-3 (IC50 10 nM), FGFR-1 (IC50 148 nM), FGFR-2 (IC50 125 nM), and FGFR-3 (IC50 68 nM). By suppressing receptor tyrosine kinase signaling pathways, Brivanib inhibits VEGF-stimulated (IC50 40 nM) and FGF-stimulated (IC50 276 nM) endothelial cell proliferation, thereby effectively blocking angiogenesis and tumor growth. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
