Biological Activity

AC480 (BMS-599626) is a potent and selective dual receptor tyrosine kinase inhibitor targeting HER1 (IC50 20 nM) and HER2 (IC50 30 nM). By blocking receptor autophosphorylation, it abrogates HER family signaling pathways and downstream cellular activation. Consequently, AC480 inhibits tumor cell proliferation and induces apoptosis in HER1- and HER2-dependent human cancer cell lines. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
